BL21-Gold(DE3)pLysS competent cells is improved version of BL21 competent cells. These high-performance competent cells provide increased transformation efficiency and produce high-quality miniprep DNA. This BL21-Gold-derived expression strain is ideal for performing protein expression studies that utilize the T7 RNA polymerase promoter to direct high-level expression. Derived from Escherichia coli B, this expression strain naturally lack the Lon protease, which can degraderecombinant proteins. In addition, these strains are engineered to be deficient for a second protease, the OmpT protein. pLysS strains express T7 lysozyme, which further suppresses basal expression of T7 RNA polymerase prior to induction, thus stabilizing pET recombinants encoding target proteins that affect cell growth and viability.
