BL21(DE3) is an exemplary host for the expression of recombinant proteins. The strain carries a deletion in the ompT gene which encodes an outer membrane protease shown to degrade recombinant products. As an E. coli B strain, it also lacks a major protease, encoded by the lon gene, which catalyzes the endoproteolytic cleavage of damaged and recombinant proteins in the cell. The BL21(DE3) is an all purpose strain for high-level protein expression and easy induction. When used with the CE6 bacteriophage, the BL21(DE3) provides the tightest control of protein expression.
